Vayne is one of the most mobile and enjoyable ADCs to play in League of Legends. Her smooth kit and quick repositioning make her extremely hard to lock down, as she will shred through any enemy in her sight. A great Vayne player can single-handedly carry games on her back, pulling off consistent montage plays.
That said, mastering Vayne is not easy and it requires good mechanics and a strong understanding of positioning. While that takes a little bit of time to practice, we’ve compiled the best builds and runes to help you out, taking information from one of the greatest Vayne pros in the world: Uzi.
Vayne ADC Build by Uzi
Vayne’s build differs from most of the traditional ADCs because she relies on her W passive to deal damage. As a result, her items are all revolved around stacking attack speed and increasing on-hit damage.
Compared to most marksmen, however, she is a lot weaker early on. This is why Vayne players usually prefer to rush Immortal Shieldbow: the shield and lifesteal are essential to survive the early-mid game.
After that, Vayne’s second core item is Guinsoo’s Rageblade. This is the real big item spike for Vayne since it applies the W passive a lot faster when the item is stacked. Instead of the usual three auto attacks, she would need two to proc W.
At this point, Vayne will start dealing significant amounts of damage, even if she falls behind. That said, the champion has to survive in order to be able to use her DPS, so Uzi goes for Blade of the Ruined King third for more healing and sustain. If he is scared of getting one shotted by a magic damage source, he builds Wit’s End instead.
To round off the build, there are a couple of choices that you can go for. If you want more attack speed, you can get Phantom Dancer. If enemies have a lot of physical damage, build Guardian Angel or Randuin’s Omen.
Unlike most ADCs, Vayne doesn’t need five damage items to be impactful: Randuin gives you both health and armor, as well as a slow to prevent enemies from chasing you. It’s the perfect item to kite people around.

Uzi uses only one rune setup for Vayne and it has Lethal Tempo as a keystone rune. The attack speed boost she gets makes her a dart-blasting cannon that can three-shot everyone in the late game. By going Precision as the primary tree, Uzi usually picks the following minor runes:
Triumph for the extra healing;
Legend: Alacrity for additional attack speed;
Coup de Grace to deal more damage to low-health targets.
The secondary rune tree is Domination, with Taste of Blood and Treasure Hunter as the go-to choices. The first gives him some early sustain for better trading while the latter grants additional gold on takedowns to get the core items faster.
